ABSTRACT The aim of this study was to evaluate the quality of Senegalia polyphylla seeds from different trees by X-ray (XR) and electric conductivity (EC) tests. Seeds from three different trees were used. The seeds were classified through radiography in: full, malformed, with small damage and empty, and were used in the germination test. For the EC test, the best volume of water (50 and 75 mL) was initially tested at 25 °C, and subsequently, the different soaking times (12, 24, 48, 72, 96 and 120 hours) using the best volume of water. Concomitantly, the germination test was performed. Tree I presented a higher percentage of full seeds in XR, higher percentage of germination and fewer leachates in the EC; tree III, however, was the opposite. The tests showed to be efficient for the quality evaluation of Senegalia polyphylla seeds, differentiating the trees.

O estudo teve como objetivo adequar a metodologia do teste de tetrazólio para estimativa da viabilidade de sementes de Eugenia uniflora (pitanga). As sementes colhidas em setembro de 2012 foram divididas em quatro lotes: recém-colhidas (lote I), armazenadas em sacos plásticos em refrigerador a 10 °C durante 15 (lote II); 30 (lote III) e 45 dias (lote IV). As sementes recém-colhidas foram submetidas aos pré-condicionamentos: embebição direta em água e em rolo de papel umedecido, ambos por 24 horas a 25 ºC e, em seguida, submetidas ou não ao corte longitudinal. Posteriormente, as sementes foram imersas em solução de tetrazólio a 0,1% por quatro horas a 30 ºC. O método de embebição e preparo mais adequados foram aplicados aos lotes, utilizando-se três concentrações de tetrazólio: 0,1; 0,5 e 1,0% e quatro tempos de coloração (2, 4, 6 e 8 horas) a 30 ºC. Os resultados obtidos pelo teste de tetrazólio foram comparados com os de germinação. A imersão direta das sementes em água por 24 horas a 25 ºC, seguida do corte longitudinal, apresentou eficiência no pré-condicionamento de sementes de pitanga. O teste de tetrazólio a 0,5% por 2 horas a 30 ºC pode ser utilizado para estimar a viabilidade tanto de sementes recém-colhidas como armazenadas.

The study aimed to adjust the methodology of the tetrazolium test to estimate seed viability of Eugenia uniflora L. (Surinam Cherry). Seeds were collected in September 2012 and divided into four lots: freshly harvested (Lot I); stored in plastic bags in a refrigerator at 10 °C for: 15 days (Lot II); 30 days (Lot III); and 45 days (Lot IV). The freshly harvested seeds were preconditioned with direct immersion in water and wet paper towel, followed or not by longitudinal cutting. The seeds were immersed in a 0.1% tetrazolium solution for 4 hours at 30 °C. Appropriate soaking and preparation methods were applied to the seeds lots using three tetrazolium concentrations: 0.1; 0.5; and 1.0%; and four preconditioning periods (2, 4, 6 and 8 hours) at 30 °C. The viability results obtained by the tetrazolium test were compared with those of the germination test. Direct immersion of seeds in water for 24 hours at 25 °C, followed by a longitudinal cut was efficient for preconditioning the seeds. Seed staining with tetrazolium solution at a concentration of 0.5% for 2 hours at 30 °C can be used to estimate the viability of freshly harvested and stored Surinam Cherry seeds.

O estudo teve como objetivo adequar a metodologia do teste de tetrazólio para avaliar a viabilidade das sementes de Eugenia involucrata DC. (cereja) e E. pyriformis Cambess. (uvaia) recém-colhidas e armazenadas. Foram empregados três lotes de sementes para ambas as espécies, sendo o I (recém-colhidas), II (armazenadas durante 15 dias) e III (armazenadas durante 30 dias). As sementes (lote I) de ambas as espécies, foram imersas em água destilada por 24 horas e submetidas a quatro métodos de preparo: sementes inteiras sem tegumento e com ¾ do tegumento; sementes cortadas longitudinalmente com tegumento e sem tegumento (imersas em solução de tetrazólio a 0,1% por 4 h). O método de preparo mais adequado foi aplicado aos demais lotes, utilizando-se três concentrações de tetrazólio: 0,075; 0,1 e 0,5% e três tempos de imersão: 2, 4 e 6 h. Os resultados do teste de tetrazólio foram comparados com os de germinação. Para a condução do teste de tetrazólio em sementes de ambas as espécies recomenda-se a embebição em água destilada por 24 horas e corte longitudinal das sementes com o tegumento. Para cereja, recomenda-se a imersão das sementes em solução de tetrazólio a 0,5% por 2 horas. Para uvaia, recomendam-se estudos adicionais.

The study aimed to adapt the tetrazolium test methodology to assess the viability of seed of Eugenia involucrata DC. (cherry) and E. pyriformis Cambess. (uvaia) freshly harvested and stored. Three lots of seeds of both species were used, being I (freshly harvested), II (stored for 15 days) and III (stored for 30 days). Seeds (lot I) of both species were immersed in distilled water for 24 hours and submitted to four preparation methods: whole seeds without seed coat and with ¾ of seed coat; longitudinally cut with and without seed coat (immersed in 0.1% tetrazolium for 4 hours). The most suitable preparation method was applied to the remaining lots, where three tetrazolium concentrations were tested: 0.075, 0.1 and 0.5% and three immersion time periods: 2, 4 and 6 hours. The tetrazolium test results were compared to the germination results. For the conduction of the tetrazolium test in seeds of both species we recommend soaking in distilled water by 24 hours and cut lengthwise on seeds with the seed coat. For cherry, the seeds should be immersed in 0.5% tetrazolium solution for 2 hours. For uvaia seeds, additional studies are recommended.

RESUMO Popularmente conhecida como sucará, Gleditschia amorphoides é uma espécie que, além do uso madeireiro, pode ser utilizada em plantios destinados à recuperação de áreas degradadas, assim, o objetivo deste trabalho foi avaliar o crescimento das mudas, ao longo do tempo, produzidas em diferentes substratos. O experimento foi realizado no viveiro da UNIOESTE em Santa Helena - PR, utilizando-se de semeadura direta em tubetes de polipropileno de 200 cm3. Foram testadas diferentes misturas como substrato, contendo Plantmax(r), resíduo de folha decomposto, serragem, cama de aviário, esterco bovino e casca de arroz carbonizada. Mensalmente, durante 180 dias, foram avaliadas altura e diâmetro do colo, sendo estes analisados segundo um delineamento de blocos casualizados, com 5 repetições de 12 mudas, em um esquema de parcelas subdivididas no tempo. No término do experimento foi avaliada a massa seca das raízes e da parte aérea, a relação entre estas variáveis e a área foliar, para 12 mudas por tratamento, seguindo um delineamento inteiramente casualizado. Determinou-se a capacidade de retenção de água, porosidade total e pH dos substratos. Todos os dados foram submetidos à análise de variância e as médias comparadas pelo teste Tukey. Em geral todos os substratos proporcionaram aumento gradativo no diâmetro e na altura das mudas. Mudas produzidas em 50 % de Plantmax(r) + 20 % de casca de arroz carbonizada + 30 % esterco bovino apresentaram maior diâmetro (4,5 mm) e altura das mudas (22,7 cm), assim como maior massa seca da raiz e da parte aérea (0,88 e 1,62 g, respectivamente), seguido de mudas produzidas em 50 % Plantmax(r) + 20 % de casca de arroz carbonizada + 20 % esterco bovino + 10 % de resíduo de folhas com 4,0 mm e 19,7 cm, para diâmetro e altura, respectivamente. Nas condições deste experimento as mudas de sucará, produzidas com 50 % de Plantmax(r) + 20 % de casca de arroz carbonizada + 30 % esterco bovino, apresentaram maior crescimento.

ABSTRACT Locally known as sucará, Gleditschia amorphoides besides being a woody species, it can be used in plantations used for the rehabilitation of degraded areas. Therefore, the objective of the current work was to evaluate the seedling growth, over time produced under different substrates. The experiment was made at the UNIOESTE nursery in Santa Helena district, in Paraná state, using direct sowing in polypropylene sized tubes of 200cm3. Different mixtures were tested as substrate, containing Plantmax(r), decomposed leaves residue, sawdust, litter, cattle manure and carbonized rice hulls. Monthly, during 180 days, evaluations of height and diameter of the lap were made, analyzed by a completely randomized design, with 5 replicates of 12 seedlings, in a subdivided plot scheme in time. At the end of the experiment, root dry mass and aerial part dry mass, the relation between these variables and the leaf area, were determined for 12 seedlings each treatment, in a completely randomized design. The water-holding capacity, substrate total porosity and pH were determined. All data were submitted to variance analysis and the means compared by the Tukey's test. In general, all substrates provided gradual increase in the diameter of the lap and seedling height. Seedlings produced in 50% of Plantmax(r)+ 20% of carbonized rice hulls + 30% of cattle manure provided bigger diameter (4,5mm) and seedlings height (22,7 cm), as well as bigger root dry mass and aerial part dry mass (0,88 and 1,62g, respectively) followed by seedlings produced in 50% of Plantmax(r)+ 20% of carbonized rice hulls + 20% of cattle manure + 10% leaf residue with 4,0mm and 19,7mm, for diameter and height respectively. In these experimental conditions, Sucará seedlings produced in 50% of Plantmax(r)+ 20% of carbonized rice hulls + 30% of cattle manure presented bigger growth.

O trabalho teve como objetivo avaliar métodos para a superação da dormência de sementes de sucará. As sementes receberam os seguintes tratamentos: 1) testemunha; 2) escarificação mecânica; 3) escarificação mecânica + água/24h; 4) escarificação mecânica + água quente/24h; 5) água quente/24h; 6) ácido sulfúrico (H2SO4)/1h; 7) H2SO4/2h; 8) H2SO4/1h + água/24h; 9) H2SO4/30min + água corrente/4h; 10) escarificação mecânica + água corrente/4h. O teste de germinação foi conduzido em rolos de papel Germitest acondicionados em câmara de germinação, à 25°C, com fotoperíodo de 12h por 26 dias. Avaliou-se também o tempo e a velocidade média de germinação. O delineamento experimental foi inteiramente casualizado, com quatro repetições de 25 sementes. O melhor desempenho germinativo foi registrado para as sementes submetidas à escarificação mecânica e química, com médias entre 76 e 98% de germinação, respectivamente, 2,17 a 2,88 dias para tempo médio de germinação e 0,46 a 0,47 sementes/dia para velocidade média de germinação, demonstrando serem estes os melhores métodos para superação da dormência de sementes dessa espécie.

The objective of this research was to evaluate methods to overcome dormancy of seeds of Gleditschia amorphoides. For dormancy's overcoming the seeds received the following treatments: 1) control; 2) mechanical scarfication 3) mechanical scarfication + water/24h; 4) mechanical scarfication + hot water/24h; 5) hot water/24h; 6) sulfuric acid (H2SO4)/1h; 7) H2SO4/2h; 8) H2SO4/1h + water/24h; 9) H2SO4/30min + flowing water/4h; 10) mechanical scarfication + flowing water/4h. Germination was done in rolls of Germitest conditioned in a germination chamber under 25°C, during 12h for 26 days. Percentage, time and average speed of germination were evaluated. The experimental design was completely casual with 10 treatments, 4 repetitions and 25 seeds. The best results were recorded for treatments with mechanical and chemical scarification with averages between 76 and 98% of germination, from 2.17 to 2.88 days for medium time for germination and 0.46 to 0.48 seeds/day for average speed of germination, demonstrating that these are the best methods to overcome dormancy of seeds.

RESUMO O presente trabalho buscou verificar os efeitos da aplicação de diferentes concentrações do ácido indolbutírico (AIB), em talco e em solução, no enraizamento de estacas caulinares de Tibouchina sellowiana (Cham.) Cogn. coletadas nas quatro estações do ano, bem como suas variações anatômicas e bioquímicas. Estacas semilenhosas foram tratadas com 0, 1500 e 3000 mg L-1 de ácido indolbutírico (AIB), em talco e solução 50% alcoólica. Após 65 dias em casa-de-vegetação, foram avaliados a porcentagem de estacas enraizadas, o número e o comprimento das raízes, a porcentagem de estacas com calos e a sobrevivência. O uso do AIB mostrou-se eficiente na indução radicial, sendo indiferente a forma em que foi aplicado. As estações mais promissoras foram a primavera (78,75%) com 3.000 mg L-1 ou mg kg-1 de AIB e o verão (63,12%) com 1.500 mg L-1 ou mg kg-1 de AIB. Das secções feitas na base das estacas, no momento da estaquia, não foi detectada nenhuma possível barreira anatômica ou diferença estrutural que promovesse variações no enraizamento. Análises bioquímicas feitas das estacas revelaram que as maiores concentrações de açúcares totais foram obtidas no inverno (83,21 mg g-1 de tecido) e no outono (72,79 mg g-1), as estações que também apresentaram as maiores concentrações de proteínas (4,69 e 3,95 mg g-1 respectivamente).

ABSTRACT The present work tried to verify the effects of application of different concentration of indolebutyric acid (IBA) in talc in solution, in the rooting of cutting of Tibouchina sellowiana (Cham.) Cogn. collected in the four seasons, as well as its anatomical and biochemical variation. The semiharwood cuttings were treated with 0, 1500 and 3000 mg L-1 indolebutyric acid (IBA), in powder and alchoolic solutions. After 65 days in a greenhouse, rooting percentage, length and number of roots, cutting with callus percentage and survival, were evaluated. The use of the IBA was efficient in the induction radicial, being indifferent the form in which it was applied. The most promising stations in the formation of the system radicial were the spring (78,75%) with 3000 mg L-1 or mg kg-1 IBA and the summer (63,12 %) with 1500 mg L-1 or mg kg-1 IBA. (82,50%) with 3000 mg kg-1 IBA. Considering the cutting in the base of the stakes, in the moment of the cutting, no possible anatomical barrier or structural difference were detected so that it should promoted variations in the answers of the rooting uring the four seasons. Biochemical analyses done from the cuttings collected in the four seasons revealed that the largest concentrations of total sugars were obtained in the winter (83,21 mg g-1) and in the autumn (72,79 mg g-1). The same stations also presented the largest protein concentrations (4,69 and 3,95 mg g-1 respectively).
